It doesn't confirm that at all... we have intel from someone who was on set that CLIFFORD is in the movie... so a cameo or appearance from Rizzo and Pepe is anything but out of the question.
I've watched it more times than I can count, too... and I'd be willing to bet that 90% of what we see of the Jason/Amy faux trailer in the beginning was filmed specifically for this trailer. I wouldn't be surprised if those parts aren't in the movie at all. I think this is a brilliant mislead...
I haven't stopped smiling for the past 30 minutes... I teared up when Kermit said, "Hi." I... have no words. We see Muppets for 45 seconds and I can already tell that this is the one we've been waiting for. Oh. My. FROG.

Unbelievably gorgeous. Just zooming into that and seeing Kermit's face... WOW. Amazing. Also... if you crop the Muppets out wide enough it makes an AMAZING...
The only way Pepe isn't in the movie is if Disney completely dropped the ball and wasn't paying attention to the production and script at all--and we all know that isn't the case. Pepe has been in the fore-front right alongside Kermit since Disney took the reins... There's no way he's going to...

Segel has said a few times that he is adamantly against having the movie in 3D... so I wouldn't worry about that. Unless Disney makes it a corporate priority to make it 3D... I doubt we'll see it this go around.
